You could go to the very rear of the truck bed where the bed liner ends and measure from left to right there to get the left and right center and then take that measurement forward. Yes, you can use a fifth wheel hitch with a bed liner. If you use a spray-on liner, you can simply install the hitch directly on top of it. If you use a hard-shell liner, you need to need to trim the liner around the hitch mounting points so that it has direct metal-to-metal contact with the truck bed.

I prefer the spray-in type of liner and would say for protection and corrosion resistance it would be the best. Also with slide-in plastic liners or similar mats, you would have to trim out the foot print of the trailer hitch rails to be able to use the 5th wheel hitch.
